CASE ALLEGATIONS: Ultragenyx is a biopharmaceutical company that focuses on the identification, acquisition, development, and commercialization of novel products for the treatment of rare and ultra-rare genetic diseases.

The Ultragenyx class action lawsuit alleges that defendants throughout the Class Period made false and/or misleading statements and/or failed to disclose that: (i) defendants created the false impression that they possessed reliable information pertaining to the effects of setrusumab on patients with variable types of Osteogenesis Imperfecta (“OI”), while also minimizing risk that patients in Ultragenyx’ Phase III Orbit study would fail to achieve a statistically significant reduction in annualized fracture rate (“AFR”), such that the second interim analysis could be performed and presented to the investing public; and (ii) in truth, Ultragenyx’ optimism in the Phase III Orbit study’s results and interim analysis benchmark were misplaced because Ultragenyx failed to convey the risk associated with basing such threshold figures on Phase II results that had no placebo control group for appropriate comparison and thus had not ruled out that the reduction in AFR from that study could merely be triggered by an increased standard of care and the placebo effect of being provided a novel treatment.

The Ultragenyx class action lawsuit further alleges that on July 9, 2025, Ultragenyx revealed that the Phase III Orbit study failed to achieve statistical significance for the second interim analysis and that Phase III Orbit and Cosmic studies would now be “progressing toward final analysis.” On this news, the price of Ultragenyx stock fell more than 25%, according to the complaint.

Then, on December 29, 2025, Ultragenyx announced that both its Phase III Orbit and Cosmic Studies had not “achieved statistical significance against the primary endpoints of reduction in annualized clinical fracture rate compared to placebo or bisphosphonates, respectively.” Ultragenyx allegedly attributed the study failure to a “low fracture rate in the placebo group” of Orbit and a trend that fell shy of statistical significance in Cosmic. On this news, the price of Ultragenyx stock fell more than 42%, according to the complaint.

CASE ALLEGATIONS: Driven Brands is an automotive services company.

Positive Data from Phase 2 CADENCE Trial Provides Definitive Proof-of-Concept for WINREVAIR™ (sotatercept-csrk) in Adults With the Syndrome of Combined Post- and Precapillary Pulmonary Hypertension and Heart Failure With Preserved Ejection Fraction

Merck’s WINREVAIR met primary endpoint of change in pulmonary vascular resistance (PVR), significantly improving the ability of blood to flow through the lungs to the heart

Totality of evidence across hemodynamic, functional, echocardiographic and clinical endpoints supports advancing development of WINREVAIR for this distinct patient population into a registrational Phase 3 study

Merck (NYSE: MRK), known as MSD outside of the United States and Canada, today announced detailed results from the Phase 2 CADENCE study, which was designed to evaluate the efficacy, safety and tolerability of two doses (0.3 mg/kg and 0.7 mg/kg) of WINREVAIR™ (sotatercept-csrk) for the treatment of adults with the syndrome of combined post- and precapillary pulmonary hypertension and heart failure with preserved ejection fraction (CpcPH-HFpEF). In this distinct patient population, WINREVAIR showed a statistically significant and clinically meaningful reduction from baseline in pulmonary vascular resistance (PVR) versus placebo (n=55) at week 24, with a 1.02 Wood units reduction for the 0.3 mg/kg dose (n=54, [95% CI, -1.81, -0.23], p=0.004) and a 0.75 Wood units reduction for the 0.7 mg/kg dose (n=55, [95% CI -1.52, 0.03], p=0.024). As noted below, important secondary endpoints explored include six-minute walk distance (6MWD), echocardiographic measures, N-terminal pro-B-type natriuretic peptide (NT-proBNP) levels and time to clinical worsening (TTCW). These late-breaking data were presented today in a late-breaking clinical trial presentation at the American College of Cardiology’s Annual Scientific Session and Expo (ACC.26), simultaneously published in Circulation and featured as part of an ACC.26 press conference.

“CpcPH-HFpEF is a distinct, identifiable and well-characterized condition that develops in people with advanced heart failure and typically impacts people who are older and have other comorbid conditions. While it is an uncommon condition and underdiagnosed, it is associated with a high morbidity and mortality rate and there are no approved treatment options specifically for CpcPH-HFpEF,” said Dr. Mardi Gomberg-Maitland, MSc, Walter G. Ross professor of clinical research, director of Pulmonary Hypertension Program, George Washington University School of Medicine and Health Sciences. “The Phase 2 CADENCE study results suggest that WINREVAIR has direct pulmonary vascular and cardiac effects in this distinct population, which may translate to clinically meaningful improvements. These proof-of-concept data provide strong rationale for further evaluation in a Phase 3 study.”

In this Phase 2 study designed to assess proof-of-concept and different doses, patients in the WINREVAIR 0.7 mg/kg arm showed an increase of 5.8 meters in 6MWD that did not reach statistical significance (95% CI, -17.3, 28.9). While subsequent secondary endpoints were not formally tested due to the prespecified hierarchical testing strategy, there was a 20.3-meter increase from baseline in 6MWD in patients treated with WINREVAIR 0.3 mg/kg (95% CI, 1.5, 39.1). Additional findings from the week 24 analysis of these secondary endpoints compared to placebo include:

  • the reduction in mean pulmonary arterial pressure (mPAP) from baseline was -9.19 for WINREVAIR 0.3 mg/kg (95% CI, -13.00, -5.38) and was -9.22 for WINREVAIR 0.7 mg/kg (95% CI, -12.97, -5.46);

  • the reduction in pulmonary arterial wedge pressure (PAWP) from baseline was -3.04 for WINREVAIR 0.3 mg/kg (95% CI, -5.77, -0.32) and -2.53 for WINREVAIR 0.7 mg/kg (95% CI, -5.33, 0.28);

  • the reduction in NT-proBNP levels from baseline was -344 pg/mL for WINREVAIR 0.3 mg/kg (95% CI, -656, -31); and -402 pg/mL for WINREVAIR 0.7 mg/kg (95% CI, -846, 42);

  • the time to first occurrence of a clinical worsening event was prolonged with WINREVAIR 0.3 mg/kg (HR: 0.18 [95% CI, 0.05, 0.62]) and WINREVAIR 0.7 mg/kg (HR: 0.59 [95% CI, 0.25, 1.36]).

The safety profile observed in CpcPH-HFpEF was generally consistent with the known safety profile for WINREVAIR in pulmonary arterial hypertension (PAH).

While both doses suggest efficacy, CADENCE results support that the 0.3 mg/kg dose may optimize the benefit-risk profile of WINREVAIR in the distinct population of CpcPH-HFpEF,” said Dr. Mahesh Patel, vice president, global clinical development, Merck Research Laboratories. “We are working with regulatory agencies to design a Phase 3 registrational study with endpoints that focus on clinical outcomes most relevant to the needs of this population, with the ultimate goal of providing the first treatment option for CpcPH-HFpEF.”

About the CADENCE study and additional results

CADENCE is a double-blind, randomized, placebo-controlled Phase 2 proof-of-concept study (NCT04945460) evaluating the efficacy, safety and tolerability of WINREVAIR versus placebo in adults with CpcPH-HFpEF. Adult patients in the trial had a diagnosis of CpcPH-HFpEF with New York Heart Association (NYHA) FC II or III. The study enrolled 164 participants, and the demographics and clinical characteristics at baseline were generally balanced. The median age was 75 years old (69-79) and 69.5% of the patients were female (n=114). 65.9% of patients had a NYHA FC III diagnosis and 34.1% had a NYHA FC II diagnosis. At baseline, 34.8% had atrial fibrillation and 46.3% had diabetes.

A total of 164 participants were randomized in a 1:1:1 ratio to one of the three treatment groups: placebo once every three weeks (Q3W) (n=55), 0.3 mg/kg WINREVAIR Q3W (n=54) and 0.7 mg/kg WINREVAIR Q3W (n=55) during the placebo-controlled treatment period. For those randomized to the 0.7 mg/kg dose of WINREVAIR, participants received a starting dose level of WINREVAIR 0.3 mg/kg for the initial three dosing visits, then escalated to the 0.7 mg/kg dose Q3W.

The CADENCE trial was designed as a proof-of-concept study with biomarkers, invasive hemodynamics, non-invasive imaging and exercise capacity. The primary endpoint is change from baseline in PVR. The study also evaluated exercise capacity, as well as echocardiographic, biomarker and clinical endpoints. Time to clinical worsening was defined by a composite endpoint of death, ≥1 hospitalization due to cardiopulmonary indication; ≥1 administration of intravenous diuretics or subcutaneous furosemide or ≥15 percent decrease from baseline in 6MWD confirmed by two tests. At baseline, the median PVR was 5.2 Wood units (4.0, 6.9), mPAP was 43 mmHg (38.0, 50.0), PAWP was 21.0 mmHg (18.0, 25.0), the median 6MWD was 273.8 meters (199.5, 343.8), and the median NT-proBNP level was 1119 pg/mL (554-2383).

Serious adverse events (SAEs) were reported in 20% of participants receiving WINREVAIR 0.3 mg/kg, 33% of participants receiving WINREVAIR 0.7 mg/kg and 22% receiving placebo. Adverse events leading to treatment discontinuation were comparable between WINREVAIR 0.3 mg/kg and placebo, with no discontinuations in either group. There were three discontinuations due to an AE and one discontinuation due to a drug-related AE in the WINREVAIR 0.7 mg/kg group. Bleeding events occurred in 26% of patients in the WINREVAIR 0.3 mg/kg group, 27% in the WINREVAIR 0.7 mg/kg group and 24% of patients in the placebo group. Adverse events leading to death occurred in one patient in the WINREVAIR 0.7 mg/kg group and two patients in the placebo group. Adverse events with an incidence of ≥10% in any group included diarrhea, fatigue, peripheral edema, influenza, nasopharyngitis, urinary tract infection, dizziness, headache and dyspnea.

About the syndrome of combined post- and precapillary pulmonary hypertension and heart failure with preserved ejection fraction (CpcPH-HFpEF)

Combined post- and precapillary pulmonary hypertension and heart failure with preserved ejection fraction (CpcPH-HFpEF) is a distinct, identifiable and well-characterized condition that develops in people with long-term or advanced heart failure. Different from Group 1 pulmonary arterial hypertension (PAH), CpcPH-HFpEF is caused by two interrelated components: pulmonary vascular disease and cardiac disease. CpcPH-HFpEF is thought to be uncommon and underdiagnosed, typically impacting people who are older and have other comorbid conditions. It is associated with a worse prognosis and higher mortality rate compared to HFpEF alone. There are no treatments specifically approved for CpcPH-HFpEF.

About WINREVAIR™ (sotatercept-csrk) for injection, for subcutaneous use, 45 mg, 60 mg

WINREVAIR is FDA-approved for the treatment of adults with pulmonary arterial hypertension (PAH, WHO Group 1 pulmonary hypertension) to improve exercise capacity and World Health Organization (WHO) functional class (FC), and reduce the risk of clinical worsening events, including hospitalization for PAH, lung transplantation and death. WINREVAIR is the first activin signaling inhibitor therapy approved to treat PAH. WINREVAIR improves the balance between pro-proliferative and anti-proliferative signaling to modulate vascular proliferation. In preclinical models, WINREVAIR induced cellular changes that were associated with thinner vessel walls, partial reversal of right ventricular remodeling and improved hemodynamics.

WINREVAIR is the subject of a licensing agreement with Bristol Myers Squibb.

Selected Safety Information for WINREVAIR

WINREVAIR may increase hemoglobin (Hgb). Severe erythrocytosis may increase the risk of thromboembolic events or hyperviscosity syndrome. Monitor Hgb before each dose for the first 5 doses, or longer if values are unstable, and periodically thereafter, to determine if dose adjustments are required.

WINREVAIR may decrease platelet count. Severe thrombocytopenia may increase the risk of bleeding. Thrombocytopenia occurred more frequently in patients also receiving prostacyclin infusion. Do not initiate treatment if platelet count is <50,000/mm3. Monitor platelets before each dose for the first 5 doses, or longer if values are unstable, and periodically thereafter to determine whether dose adjustments are required.

In clinical studies, serious bleeding (e.g., gastrointestinal, intracranial hemorrhage) was reported in 4% vs 1% (STELLAR) and 7% vs 5% (ZENITH) of patients taking WINREVAIR vs placebo, respectively. Patients with serious bleeding were more likely to be on prostacyclin background therapy and/or antithrombotic agents, or have low platelet counts. Advise patients about signs and symptoms of blood loss. Evaluate and treat bleeding accordingly. Do not administer WINREVAIR if the patient is experiencing serious bleeding.

WINREVAIR may cause fetal harm when administered to a pregnant woman. Advise pregnant women of the potential risk to a fetus. Advise females of reproductive potential to use an effective method of contraception during treatment with WINREVAIR and for at least 4 months after the final dose. Pregnancy testing is recommended for females of reproductive potential before starting WINREVAIR treatment.

Based on findings in animals, WINREVAIR may impair female and male fertility. Advise patients on the potential effects on fertility.

The most common adverse reactions (≥10% for WINREVAIR and at least 5% more than placebo) occurring in the STELLAR Phase 3 clinical trial were headache (24.5% vs 17.5%), epistaxis (22.1% vs 1.9%), rash (20.2% vs 8.1%), telangiectasia (16.6% vs 4.4%), diarrhea (15.3% vs 10.0%), dizziness (14.7% vs 6.3%) and erythema (13.5% vs 3.1%). The most common adverse reactions in the ZENITH trial were infections (67.4% vs 44.2%), epistaxis (45.3% vs 9.3%), diarrhea (25.6 % vs 17.4%), telangiectasia (25.6 % vs 3.5%), increased hemoglobin (15.1% vs 1.2%), rash (10.5% vs 4.7%), erythema (10.5% vs 3.5%) and gingival bleeding (10.5% vs 2.3%).

Because of the potential for serious adverse reactions in the breastfed child, advise patients that breastfeeding is not recommended during treatment with WINREVAIR, and for 4 months after the final dose.

Bristol Myers Squibb Presents Positive Results from Phase 3 SCOUT-HCM Trial Demonstrating Efficacy and Safety of Camzyos (mavacamten) in Adolescents with Symptomatic Obstructive Hypertrophic Cardiomyopathy (oHCM)

Trial met its primary endpoint, demonstrating clinically meaningful and statistically significant reduction in Valsalva left ventricular outflow tract (LVOT) gradient at Week 28

Results demonstrate the potential for Camzyos to be the first targeted pharmacological therapy for the treatment of oHCM in adolescents

Safety profile of Camzyos in adolescents was similar to established profile in adults, with no new safety signals and no patients experiencing left ventricular ejection fraction (LVEF) of <50%

PRINCETON, N.J.–(BUSINESS WIRE)–Bristol Myers Squibb (NYSE: BMY) today announced positive data from the Phase 3 SCOUT-HCM trial of Camzyos (mavacamten), the first study of a cardiac myosin inhibitor (CMI) in adolescents (ages 12 years to <18 years) with symptomatic obstructive hypertrophic cardiomyopathy (oHCM). The SCOUT-HCM trial met its primary endpoint, demonstrating a clinically meaningful and statistically significant reduction from baseline in Valsalva left ventricular outflow tract (LVOT) gradient at Week 28 with Camzyos versus placebo, with a significant least-squares (LS) mean difference (95% CI) at Week 28 of −48.0 (−67.7, −28.3) mm Hg; P < 0.0001.

Additionally, Camzyos showed meaningful improvement over placebo in multiple secondary endpoints at 28 weeks, and similar safety findings were observed in the Camzyos and placebo groups. The data are being presented as a late-breaking clinical trial presentation at the American College of Cardiology’s (ACC) Annual Scientific Session & Expo 2026, with simultaneous publication in The New England Journal of Medicine.

“Pediatric HCM is a rare cardiac disorder that is associated with severe, sometimes life-threatening, symptoms,” said Joseph Rossano, MD, Principal Investigator of SCOUT-HCM and Chief of the Division of Cardiology at Children’s Hospital of Philadelphia. “With no approved therapies for pediatric patients with oHCM and current recommendations for pharmacological therapy primarily extrapolated from evidence obtained from adult studies, the positive results of this trial represent a significant advance in the field of pediatric cardiology and the potential for a meaningful new therapy for adolescent patients if approved by the FDA.”

SCOUT-HCM evaluated Camzyos compared to placebo in 44 patients aged 12 to <18 years old with symptomatic oHCM and New York Heart Association (NYHA) class II-III symptoms over a 28-week period. In addition to meeting the primary endpoint of reduction of Valsalva LVOT gradient, Camzyos showed meaningful improvements over placebo in LV obstruction, diastolic function, maximal left ventricular wall thickness, NYHA class, and mitral valve dysfunction at 28 weeks.

Improvements were seen in resting and post-exercise LVOT gradients with Camzyos versus placebo, with an LS mean difference of −47.0 mm Hg (−62.7, −31.4); nominal p < 0.0001, and −41.7 mm Hg (−59.7, −23.7); nominal p < 0.0001, respectively. The primary endpoint was supported by the proportion of patients who achieved reduction in maximal LVOT gradient (resting or Valsalva) to <30 mm Hg, from baseline to 28 weeks.

Additionally, positive structural changes were observed with Camzyos versus placebo, as shown by improvement in maximal LV wall thickness LS mean difference (95% CI) of −1.8 (−3.4, −0.2) mm; nominal p = 0.0269 and average E/e’ ratio (ratio between early mitral inflow velocity and mitral annular early diastolic velocity) of LS mean difference (95% CI) of −3.4 (−5.1, −1.6); nominal p = 0.0002.

Similar safety findings were observed in the Camzyos and placebo groups, including treatment-emergent adverse events (TEAEs) (18 versus 17 participants experiencing at least 1 respectively), treatment-related TEAEs (2 versus 3, respectively), treatment-emergent serious adverse events (TESAEs) (2 for both groups), and treatment-related TESAEs (1 versus 0, respectively). During the 28-week study period, there were no TEAE‑related treatment discontinuations or deaths, no cases of atrial fibrillation or symptomatic heart failure, and no patient experienced a left ventricular ejection fraction (LVEF) of <50%. Overall, these findings demonstrate that the safety profile of Camzyos treatment in symptomatic adolescents with oHCM was similar to that reported in adults, with no new safety signals identified.

The 28-week active treatment period of SCOUT-HCM is ongoing, and Bristol Myers Squibb will work with key investigators to present the 56-week data at an upcoming medical congress.

About the Phase 3 SCOUT-HCM Trial

SCOUT-HCM (NCT06253221) is a Phase 3 randomized, double-blind, placebo-controlled, international trial that enrolled 44 adolescent patients (12 to <18 years old) with symptomatic oHCM. The trial includes three treatment periods totaling up to 200 weeks: a 28-week placebo-controlled period, followed by a 28-week active-treatment period (when patients randomized to placebo crossed over to Camzyos), and an open-label long-term extension period for up to 144 weeks.

The primary endpoint is change from baseline to Week 28 in Valsalva LVOT gradient. Secondary endpoints include efficacy parameters of resting and post-exercise LVOT gradients, peak oxygen consumption, symptoms and health status, plus safety and pharmacokinetic parameters.

About Obstructive Hypertrophic Cardiomyopathy (oHCM) in Adolescent Patients

Hypertrophic cardiomyopathy (HCM) is a primary cardiac disorder that may result from known or suspected genetic defects in sarcomeric proteins of the cardiac myocyte or be due to unknown reasons (idiopathic). Adolescents with obstructive HCM suffer substantial morbidity largely related to reduced exertional tolerance. Though available treatments can lead to improvement in symptoms, they have significant limitations for adolescent patients (e.g., side effects from beta-blockers and risks of invasive procedures).

About CAMZYOS® (mavacamten)

CAMZYOS® (mavacamten) is the most extensively studied cardiac myosin inhibitor (CMI), approved by regulatory bodies in more than 60 countries and regions across five continents worldwide. CAMZYOS U.S. IMPORTANT SAFETY INFORMATION

WARNING: RISK OF HEART FAILURE

CAMZYOS reduces left ventricular ejection fraction (LVEF) and can cause heart failure due to systolic dysfunction.

Echocardiogram assessments of LVEF are required prior to and during treatment with CAMZYOS. Initiation of CAMZYOS in patients with LVEF <55% is not recommended. Interrupt CAMZYOS if LVEF is <50% at any visit or if the patient experiences heart failure symptoms or worsening clinical status.

Concomitant use of CAMZYOS with certain cytochrome P450 inhibitors or discontinuation of certain cytochrome P450 inducers may increase the risk of heart failure due to systolic dysfunction; therefore, the use of CAMZYOS is contraindicated with the following:

  • Strong CYP2C19 inhibitors
  • Moderate to strong CYP2C19 inducers or moderate to strong CYP3A4 inducers

Because of the risk of heart failure due to systolic dysfunction, CAMZYOS is available only through a restricted program under a Risk Evaluation and Mitigation Strategy (REMS) called CAMZYOS REMS PROGRAM.

CONTRAINDICATIONS

CAMZYOS is contraindicated with concomitant use of:

  • Strong CYP2C19 inhibitors

  • Moderate to strong CYP2C19 inducers or moderate to strong CYP3A4 inducers

WARNINGS AND PRECAUTIONS

Heart Failure

CAMZYOS reduces systolic contraction and can cause heart failure or significantly reduce ventricular function. Patients who experience a serious intercurrent illness (e.g., serious infection) or arrhythmia (e.g., atrial fibrillation or other uncontrolled tachyarrhythmia) are at greater risk of developing systolic dysfunction and heart failure.

Assess the patient’s clinical status and LVEF prior to and regularly during treatment and adjust the CAMZYOS dose accordingly. New or worsening arrhythmia, dyspnea, chest pain, fatigue, palpitations, leg edema, or elevations in N-terminal pro-B-type natriuretic peptide (NT-proBNP) may be signs and symptoms of heart failure and should also prompt an evaluation of cardiac function.

Asymptomatic LVEF reduction, intercurrent illnesses, and arrhythmias require additional dosing considerations.

Initiation of CAMZYOS in patients with LVEF <55% is not recommended. Avoid concomitant use of CAMZYOS in patients on disopyramide, ranolazine, verapamil with a beta blocker, or diltiazem with a beta blocker as these medications and combinations increase the risk of left ventricular systolic dysfunction and heart failure symptoms and clinical experience is limited.

CYP450 Drug Interactions Leading to Heart Failure or Loss of Effectiveness

CAMZYOS is primarily metabolized by CYP2C19 and CYP3A4 enzymes. Concomitant use of CAMZYOS and drugs that interact with these enzymes may lead to life-threatening drug interactions such as heart failure or loss of effectiveness.

Advise patients of the potential for drug interactions, including with over-the-counter medications (such as omeprazole, esomeprazole, or cimetidine). Advise patients to inform their healthcare provider of all concomitant products prior to and during CAMZYOS treatment.

CAMZYOS Risk Evaluation and Mitigation Strategy (REMS) Program

CAMZYOS is only available through a restricted program called the CAMZYOS REMS Program because of the risk of heart failure due to systolic dysfunction. Notable requirements of the CAMZYOS REMS Program include the following:

  • Prescribers must be certified by enrolling in the REMS Program

  • Patients must enroll in the REMS Program and comply with ongoing monitoring requirements

  • Pharmacies must be certified by enrolling in the REMS Program and must only dispense to patients who are authorized to receive CAMZYOS

  • Wholesalers and distributors must only distribute to certified pharmacies

Embryo-Fetal Toxicity

CAMZYOS may cause fetal toxicity when administered to a pregnant female, based on findings in animal studies. Confirm absence of pregnancy in females of reproductive potential prior to treatment and advise patients to use effective contraception during treatment with CAMZYOS and for 4 months after the last dose. Combined hormonal contraceptives (CHCs) containing a combination of ethinyl estradiol and norethindrone may be used with CAMZYOS. However, CAMZYOS may reduce the effectiveness of certain other CHCs. If these CHCs are used, advise patients to add nonhormonal contraception (such as condoms) during concomitant use and for 4 months after the last dose of CAMZYOS.

ADVERSE REACTIONS

In the EXPLORER-HCM trial, adverse reactions occurring in >5% of patients and more commonly in the CAMZYOS group than in the placebo group were dizziness (27% vs 18%) and syncope (6% vs 2%). There were no new adverse reactions identified in VALOR-HCM.

Effects on Systolic Function

In the EXPLORER-HCM trial, mean (SD) resting LVEF was 74% (6) at baseline in both treatment groups. Mean (SD) absolute change from baseline in LVEF was -4% (8) in the CAMZYOS group and 0% (7) in the placebo group over the 30-week treatment period. At Week 38, following an 8-week interruption of trial drug, mean LVEF was similar to baseline for both treatment groups. In the EXPLORER-HCM trial, 7 (6%) patients in the CAMZYOS group and 2 (2%) patients in the placebo group experienced reversible reductions in LVEF <50% (median 48%: range 35-49%) while on treatment. In all 7 patients treated with CAMZYOS, LVEF recovered following interruption of CAMZYOS.

DRUG INTERACTIONS

Potential for Other Drugs to Affect Plasma Concentrations of CAMZYOS

CAMZYOS is primarily metabolized by CYP2C19 and to a lesser extent by CYP3A4 and CYP2C9. Inducers and inhibitors of CYP2C19 and moderate to strong inhibitors or inducers of CYP3A4 may affect the exposures of CAMZYOS.

Impact of Other Drugs on CAMZYOS:

  • Strong CYP2C19 Inhibitors: Concomitant use increases CAMZYOS exposure, which may increase the risk of heart failure due to systolic dysfunction. Concomitant use is contraindicated.
  • Moderate to Strong CYP2C19 Inducers or Moderate to Strong CYP3A4 Inducers: Concomitant use decreases CAMZYOS exposure, which may reduce CAMZYOS’ efficacy. The risk of heart failure due to systolic dysfunction may increase with discontinuation of these inducers as the levels of induced enzyme normalizes. Concomitant use is contraindicated.
  • Weak CYP2C19 Inhibitors or Moderate CYP3A4 Inhibitors: Concomitant use with a weak CYP2C19 inhibitor or a moderate CYP3A4 inhibitor increases CAMZYOS exposure, which may increase the risk of adverse drug reactions. Initiate CAMZYOS at the recommended starting dose of 5 mg orally once daily in patients who are on stable therapy with a weak CYP2C19 inhibitor or a moderate CYP3A4 inhibitor. Reduce dose of CAMZYOS by one level (ie, 15 to 10 mg, 10 to 5 mg, or 5 to 2.5 mg) in patients who are on CAMZYOS treatment and intend to initiate a weak CYP2C19 inhibitor or a moderate CYP3A4 inhibitor. Schedule clinical and echocardiographic assessment 4 weeks after inhibitor initiation, and do not up-titrate CAMZYOS until 12 weeks after inhibitor initiation. Avoid initiation of concomitant weak CYP2C19 and moderate CYP3A4 inhibitors in patients who are on stable treatment with 2.5 mg of CAMZYOS because a lower dose is not available. For short-term use (eg, 1 week), interrupt CAMZYOS for the duration of treatment with a weak inhibitor of CYP2C19 or a moderate inhibitor of CYP3A4. CAMZYOS may be reinitiated at the previous dose immediately on discontinuation of concomitant therapy.
  • Moderate CYP2C19 Inhibitors or Strong CYP3A4 Inhibitors: Concomitant use with a moderate CYP2C19 inhibitor or strong CYP3A4 inhibitor increases CAMZYOS exposure, which may increase the risk of adverse drug reactions. Discontinuing use of a moderate CYP2C19 inhibitor or strong CYP3A4 inhibitor after long-term concomitant use may decrease CAMZYOS exposure, which may reduce CAMZYOS’ efficacy. Initiate CAMZYOS at a starting dosage of 2.5 mg orally once daily in patients who are on a stable therapy with a moderate CYP2C19 inhibitor or a strong CYP3A4 inhibitor. Reduce dose of CAMZYOS by one level (ie, 15 to 10 mg, 10 to 5 mg, or 5 to 2.5 mg) in patients who are on CAMZYOS and intend to initiate a moderate CYP2C19 inhibitor or a strong CYP3A4 inhibitor. Avoid initiation of concomitant moderate CYP2C19 and strong CYP3A4 inhibitors in patients who are on a stable treatment with 2.5 mg of CAMZYOS because a lower dose is not available. An increase in dose of CAMZYOS may be needed if the moderate inhibitor of CYP2C19 or strong inhibitor of CYP3A4 is discontinued after long-term concomitant use. Monitor for new or worsening symptoms. For short-term use (ie, when CAMZYOS dose modification is not feasible), interrupt CAMZYOS for the duration of treatment with a moderate inhibitor of CYP2C19 or a strong inhibitor of CYP3A4. CAMZYOS may be reinitiated at the previous dose immediately on discontinuation of concomitant therapy.

Potential for CAMZYOS to Affect Plasma Concentrations of Other Drugs

CAMZYOS is an inducer of CYP3A4, CYP2C9, and CYP2C19. Concomitant use with CYP3A4, CYP2C9, or CYP2C19 substrates may reduce plasma concentration of these drugs. Closely monitor when CAMZYOS is used with concomitant CYP3A4, CYP2C9, or CYP2C19 substrates unless otherwise recommended in the Prescribing Information.

Certain Combined Hormonal Contraceptives (CHCs): Progestin and ethinyl estradiol are CYP3A4 substrates. Concomitant use of CAMZYOS may decrease exposures of certain progestins, which may lead to contraceptive failure. CHCs containing a combination of ethinyl estradiol and norethindrone may be used with CAMZYOS, but if other CHCs are used, advise patients to add nonhormonal contraception (such as condoms) or use an alternative contraceptive method that is not affected by CYP450 enzyme induction (eg, intrauterine system) during concomitant use and for 4 months after the last dose of CAMZYOS.

Drugs That Reduce Cardiac Contractility

Expect additive negative inotropic effects of CAMZYOS and other drugs that reduce cardiac contractility. Avoid concomitant use of CAMZYOS in patients on disopyramide, ranolazine, verapamil with a beta blocker, or diltiazem with a beta blocker as these medications and combinations increase the risk of left ventricular systolic dysfunction and heart failure symptoms and clinical experience is limited.

If concomitant therapy with a negative inotrope is initiated, or if the dose of a negative inotrope is increased, monitor LVEF closely until stable doses and clinical response have been achieved.

SPECIFIC POPULATIONS

Pregnancy

Based on animal data, CAMZYOS may cause fetal harm when administered to a pregnant female. Advise pregnant females about the potential risk to the fetus with maternal exposure to CAMZYOS during pregnancy. There is a pregnancy safety study for CAMZYOS. If CAMZYOS is administered during pregnancy, or if a patient becomes pregnant while receiving CAMZYOS or within 4 months after the last dose of CAMZYOS, healthcare providers should report CAMZYOS exposure by contacting Bristol Myers Squibb at 1-800-721-5072 or www.bms.com.

Lactation

The presence of CAMZYOS in human or animal milk, the drug’s effects on the breastfed infant, or the effects on milk production are unknown. The developmental and health benefits of breastfeeding should be considered along with the mother’s clinical need for CAMZYOS and any potential adverse effects on the breastfed child from CAMZYOS or from the underlying maternal condition.

Females and Males of Reproductive Potential

Confirm absence of pregnancy in females of reproductive potential prior to initiation of CAMZYOS. Advise females of reproductive potential to use effective contraception during treatment with CAMZYOS and for 4 months after the last dose. CHCs containing a combination of ethinyl estradiol and norethindrone may be used with CAMZYOS. However, CAMZYOS may reduce the effectiveness of certain other CHCs. ZUG, Switzerland, March 28, 2026 – MoonLake Immunotherapeutics (NASDAQ: MLTX) (MoonLake or the Company), a clinical-stage biotechnology company focused on creating next-level therapies for inflammatory diseases, today announces long-term Week 40 results of the Phase 3 VELA-1 and VELA-2 clinical trials of its registrational global program in patients with moderate-to-severe HS and confirms the presentation of the data at the 2026 American Academy of Dermatology (AAD) Annual Meeting later today.

The Phase 3 VELA program in adults with moderate-to-severe HS used the higher clinical response level of HS Clinical Response (HiSCR) 75 as the primary endpoint, which defines a response as an at least 75% reduction in abscess and inflammatory nodule count, with no increase from baseline in abscess or draining tunnel count. Key secondary endpoints included the change from baseline in the HS Quality of Life score (HiSQOL) as well as other scores that reflect the evolving needs of HS patients, treating physicians and regulators. These included the percentage of participants achieving at least a 55% reduction in the International HS Severity Scoring System (IHS4-55), the percentage of participants achieving at least a 3-point improvement from baseline in the worst skin pain Numerical Rating Scale (NRS) among participants with a baseline score of at least 3 points, and the percentage of patients achieving a Dermatology Quality of Life Index (DLQI) total score improvement of ≥4 (minimal clinically important difference), among participants with a baseline DLQI ≥4. A total of 838 patients were enrolled across both trials. Following the primary endpoint at Week 16, patients in the placebo arm were switched to receive SLK treatment for the remaining duration of the trial until Week 52. Patients originally randomized to the SLK arm continued to receive SLK at the monthly 120mg maintenance dose. All patients have completed Week 40 and discontinuation rates were at the low end to those observed in other pivotal HS trials. After Week 52, patients have the opportunity to switch into a two-year open-label extension trial.

SLK demonstrated continued improvement of clinical scores and Patient Reported Outcomes (PROs) at Week 40. Across both VELA-1 and VELA-2, 62% of patients treated with SLK achieved HiSCR75 response and up to 32% of patients achieved HiSCR100 response at Week 40 (as observed). The results were consistent across both trials (VELA-1: 61.9% HiSCR75, 32.4% HiSCR100; VELA-2: 61.8% HiSCR75, 28.1% HiSCR100; as observed). Up to 77% of patients achieved an IHS-4 55 response (VELA-1: 74.3%, VELA-2: 77.4%; as observed) and up to 25% of patients achieved inflammatory remission at Week 40 (A100 + DT100 + N100; VELA-1: 25.2%, VELA-2: 21.1%; as observed).

With 62% of SLK patients reaching a HiSCR75 response already at Week 40, SLK sets a new standard in long-term lesion control of HS. Approved IL-17A and IL17A & F inhibitors reported HiSCR75 response rates of approximately 40% and 60%, respectively, at the 1-year mark of their respective pivotal trials (approved doses, pooled analysis across pivotal trials).

The strong long-term clinical responses were accompanied by continued improvement in PROs, which reflect critical quality of life outcomes for patients with HS. Patients treated with SLK showed a significant improvement of HiSQOL score at Week 40 with a -11.8 change from baseline in VELA-1, and -12.4 change from baseline in VELA-2 (mean HiSQOL at baseline of 26.5 (VELA-1) and 28.0 (VELA-2); as observed). While 59% of patients were within the “very severe” category on the HiSQOL severity score at baseline, 63% of patients were in the “mild / none” category at Week 40 (as observed, pooled). Improvements versus baseline in HiSQOL-Mini items ranged from 41% (pain), to 54% (walking, getting dressed) to 62% (down or depressed) at Week 40 (in patients with baseline >0, as observed, pooled). In addition, up to 43% of patients experienced a marked reduction of pain, as measured by an at least 3-point improvement from baseline in the worst skin pain NRS (VELA-1: 43.2%, VELA-2: 37.2%; as observed, baseline score of at least three points). 65% of patients achieved a meaningful (at least four points from baseline) improvement in DLQI (VELA-1: 64.6%, VELA-2: 65.3%; as observed, baseline DLQI of at least four points).

The safety profile of SLK in the VELA clinical trials remains consistent over time, with no new safety signals detected.

About Sonelokimab

Sonelokimab (M1095) is an investigational ~40 kDa humanized Nanobody® consisting of three VHHs covalently linked by flexible glycine-serine spacers. With two domains, sonelokimab selectively binds with high affinity to IL-17A and IL-17F, thereby inhibiting the IL-17A/A, IL-17A/F, and IL-17F/F dimers. A third central domain binds to human albumin, facilitating further enrichment of sonelokimab at sites of inflammatory edema.

Sonelokimab is being assessed in two lead indications, hidradenitis suppurativa (HS) and psoriatic arthritis (PsA), and the Company is pursuing other indications in dermatology and rheumatology, including adolescent HS, palmoplantar pustulosis (PPP) and axial spondyloarthritis (axSpA).

For adults with HS, sonelokimab is being assessed in two identical Phase 3 trials, the VELA-1 and VELA-2 trials, using the higher clinical response level of HS Clinical Response (HiSCR) 75 as the primary endpoint, which defines a response as an at least 75% reduction in abscess and inflammatory nodule count, with no increase from baseline in abscess or draining tunnel count. In September 2025, the primary endpoint data from the VELA-1 and VELA-2 clinical trials were announced. In the combined VELA program, patients treated with SLK experienced a clinically meaningful and statistically significant improvement across all primary and key secondary endpoints using both pre-specified strategies (p<0.001). In VELA-1, SLK achieved statistical significance for all primary and key secondary endpoints using both pre-specified strategies (HiSCR75, delta to placebo of 17%, p<0.001). In VELA-2, intercurrent events in the higher-than-expected placebo arm precluded the study from achieving statistical significance in the week 16 primary endpoint using the composite strategy (HiSCR75, delta to placebo of 9%, p=0.053). From week 16, all patients are expected to continue to receive the 120mg dose of SLK through to 48 weeks, with a last assessment planned at week 52, followed by an open-label extension for up to two years. The safety profile of sonelokimab in the VELA trials was consistent with previous trials with no new safety signals detected.

Sonelokimab is currently undergoing evaluation in the VELA-TEEN Phase 3 trial, which is the first clinical study specifically focused on adolescent patients with moderate-to-severe HS.

For PsA, sonelokimab is being assessed in the Phase 3 trials, IZAR-1 and IZAR-2, following the announcement in March 2024 of the full dataset from the global Phase 2 ARGO trial (M1095-PSA-201) evaluating the efficacy and safety of the Nanobody® sonelokimab over 24 weeks in patients with active PsA. Significant improvements were observed across all key outcomes, including approximately 60% of patients treated with sonelokimab achieving an American College of Rheumatology (ACR) 50 response and Minimal Disease Activity (MDA) at week 24. This followed the positive top-line results in November 2023, where the trial met its primary endpoint with a statistically significant greater proportion of patients treated with either sonelokimab 60mg or 120mg (with induction) achieving an ACR50 response compared to those on placebo at week 12. All key secondary endpoints in the trial were met for the 60mg and 120mg doses with induction. The safety profile of sonelokimab in the ARGO trial was consistent with previous trials with no new safety signals detected.

Sonelokimab is also being assessed in PPP, a debilitating inflammatory skin condition affecting a significant number of patients, including in the completed Phase 2 LEDA program. In the Phase 2 LEDA clinical trial in PPP, SLK demonstrated clinically meaningful and statistically significant benefit. Patients treated with SLK achieved a mean percent change from baseline in the Palmoplantar Pustular Psoriasis Area and Severity Index (PPPASI) of 64% at week 16, and 39% of patients achieved a ≥75% reduction in the PPPASI (PPPASI75), suggesting that SLK could provide clinically meaningful improvements in this disease for which there are currently no approved therapies. The safety profile of SLK in the LEDA trial was consistent with previous trials with no new safety signals detected. 

Additionally, Sonelokimab is being assessed in the ongoing Phase 2 S-OLARIS and P-OLARIS trials for active axSpA and PsA, respectively. Both trials feature an innovative design complementing traditional clinical outcomes with cellular imaging techniques.

Sonelokimab has also been assessed in a randomized, placebo-controlled third-party Phase 2b trial (NCT03384745) in 313 patients with moderate-to-severe plaque-type psoriasis. High threshold clinical responses (Investigator’s Global Assessment Score 0 or 1, and Psoriasis Area and Severity Index 90/100) were observed in patients with moderate-to-severe plaque-type psoriasis. Sonelokimab generally presented a safety profile similar to the active control, secukinumab (Papp KA, et al. Lancet. 2021; 397:1564-1575).

In an earlier third-party Phase 1 trial in patients with moderate-to-severe plaque-type psoriasis, sonelokimab decreased (to normal skin levels) the cutaneous gene expression of pro-inflammatory cytokines and chemokines (Svecova D. J Am Acad Dermatol. 2019; 81:196–203).

About the VELA program

The Phase 3 VELA program has enrolled over 800 patients across VELA-1 and VELA-2. Both global, randomized, double-blind, and placebo-controlled trials are identical in design evaluating the efficacy and safety of the Nanobody® sonelokimab, administered subcutaneously, in adult patients with active moderate-to-severe hidradenitis suppurativa. Similar to the design of the landmark Phase 2 MIRA trial, the primary endpoint is the percentage of participants achieving Hidradenitis Suppurativa Clinical Response (HiSCR) 75, defined as a ≥75% reduction in total abscess and inflammatory nodule (AN) count with no increase in abscess or draining tunnel count relative to baseline. The trials also evaluate a number of secondary endpoints, including the proportion of patients achieving HiSCR50, the change from baseline in International Hidradenitis Suppurativa Severity Score System (IHS4), the proportion of patients achieving a Dermatology Life Quality Index (DLQI) total reduction of ≥4, the proportion of patients achieving at least 50% reduction from baseline in Numerical Rating Scale (NRS50) in the Patient’s Global Assessment of Skin Pain (PGA Skin Pain) and complete resolution of Draining Tunnels (DT100). The VELA protocols and statistical analysis plans were prepared in accordance with regulatory agency advice and include two analysis strategies. The composite strategy for the VELA trials (also referred to as the primary estimand) is the primary statistical analysis. The protocol specifies the treatment policy strategy as the alternative method of handling intercurrent events to test the robustness of the VELA data. About the MIRA trial

The MIRA trial (M1095-HS-201) is a global, randomized, double-blind, placebo-controlled Phase 2 trial to evaluate the efficacy and safety of the Nanobody® sonelokimab, administered subcutaneously, in the treatment of adult patients with active moderate-to-severe hidradenitis suppurativa. The trial recruited 234 patients, with the aim to evaluate two different doses of sonelokimab (120mg and 240mg) with placebo control and adalimumab as an active reference arm. The primary endpoint of the trial is the percentage of participants achieving Hidradenitis Suppurativa Clinical Response 75 (HiSCR75), defined as a ≥75% reduction in total abscess and inflammatory nodule (AN) count with no increase in abscess or draining tunnel count relative to baseline. About the VELA-TEEN trial

The Phase 3 VELA-TEEN trial is an open-label, single-arm trial designed to evaluate sonelokimab 120mg administered subcutaneously once every two weeks (Q2W) until week six and once every four weeks (Q4W) from week eight onwards. The trial aims to enroll 30-35 adolescents, aged 12-17, with moderate-to-severe hidradenitis suppurativa, from U.S. sites experienced in clinical trials and pediatric dermatology. The primary trial phase will be 24 weeks with a primary endpoint evaluating the pharmacokinetics, safety, and tolerability of sonelokimab. About Hidradenitis Suppurativa

Hidradenitis suppurativa (HS) is a severely debilitating chronic skin condition resulting in irreversible tissue destruction. HS manifests as painful inflammatory skin lesions, typically around the armpits, groin, and buttocks. Over time, uncontrolled and inadequately treated inflammation can result in irreversible tissue destruction and scarring. The disease affects an estimated 2% of the population, with three times more females affected than males. About Psoriatic Arthritis

Psoriatic arthritis (PsA) is a chronic, progressive and complex inflammatory disease that manifests across multiple domains, leading to substantial functional impairment and decreased quality of life. The clinical features of PsA are diverse, comprising both musculoskeletal (peripheral arthritis, spondylitis, dactylitis, and enthesitis) and non-musculoskeletal (skin and nail disease) domains. PsA occurs in up to 30% of patients with psoriasis, most commonly those aged between 30 and 60 years. About the S-OLARIS trial

The S-OLARIS trial (M1095-axSpA-201) is a Phase 2 trial designed to evaluate the efficacy and safety of sonelokimab 60mg administered subcutaneously in adult patients with active axial spondyloarthritis (axSpA). The trial recruited 26 patients. The primary endpoint is the change from baseline (CfB) in 18F-NaF SUVmax signals at week 12 in the sacroiliac joints and spine as detected by PET. Throughout the trial, several other endpoints will be assessed including established clinical disease activity outcomes (e.g., ASAS), scores related to physical function, spinal mobility, and enthesitis as well as patient reported outcomes. About Axial Spondyloarthritis

Axial Spondyloarthritis (axSpA) typically impacts young people, with diagnosis based on chronic inflammatory back pain lasting more than three months with onset under 45 years of age. Advanced disease can lead to progressive and pathologic bone formation and joint fusion, severely limiting spinal mobility. Global reported prevalence of axSpA ranges from 0.5% to 1.5%. AxSpA can be categorized by disease progression into two subtypes: non-radiographic axSpA and ankylosing spondylitis (AS), also known as radiographic axSpA, which is diagnosed based on radiographic evidence of structural changes to the sacroiliac joints. Patients with axSpA experience fatigue, persistent morning stiffness, and pain that worsens at night and can disrupt sleep. About Palmoplantar Pustulosis

Palmoplantar Pustulosis (PPP) is characterized by the development of blister-like pustules within erythematous, scaly plaques on the palms and the soles of the feet. PPP typically develops in adulthood, more frequently impacts females. Biogen Announces Second Positive Phase 2 Litifilimab Trial in Cutaneous Lupus Erythematosus at 2026 American Academy of Dermatology Annual Meeting, Showing a Significant Reduction in Skin Disease Activity

  • In the Phase 2 part of the AMETHYST study, litifilimab met the primary endpoint of reduction of disease activity in people living with CLE at Week 16, with more litifilimab participants achieving clear / almost clear skin

CAMBRIDGE, Mass., March 28, 2026 (GLOBE NEWSWIRE) — Biogen Inc. (Nasdaq: BIIB) announced positive results from the Phase 2 part of the AMETHYST Phase 2/3 study (Part A) of litifilimab in people living with cutaneous lupus erythematosus (CLE). Litifilimab is the first humanized IgG1 monoclonal antibody (mAb) targeting blood dendritic cell antigen 2 (BDCA2), which has the potential to become the first innovative therapy approved for CLE in 70 years. Part A of AMETHYST evaluated the efficacy and safety of litifilimab through week 24, with reductions in skin disease activity reported across several measures. Results are consistent with the earlier positive Phase 2 LILAC study reported in The New England Journal of Medicine. Phase 2 results from LILAC and AMETHYST supported litifilimab’s recently granted U.S. Food and Drug Administration (FDA) Breakthrough Therapy Designation. Results are being presented today at the American Academy of Dermatology (AAD) Annual Meeting.

CLE is a complex, heterogenous disease that affects millions of people around the world. Today there are no targeted therapies approved to treat CLE.

AMETHYST is an ongoing seamless two-part, multicenter, double-blind, placebo controlled, randomized study to evaluate the efficacy and safety of litifilimab compared to placebo in participants with a variety of CLE severities. Participants are randomized to receive subcutaneous treatment with litifilimab with standard of care (SoC) or placebo every four weeks in addition to SoC. All participants will receive litifilimab during the 28-week extended treatment period from Weeks 24 to 48. Results reported here are from the double-blind, placebo-controlled Phase 2 (Part A) portion of AMETHYST, Week 0 to 24. In the study, 74% of participants are women and 33% are non-white. This demographic distribution is consistent with the epidemiology of CLE which disproportionately affects women and diverse ethno-racial groups. The Phase 3 part of the study is ongoing and results remain blinded.  

AMETHYST Part A met its primary endpoint with litifilimab demonstrating a statistically significant 11.8% higher reduction in disease activity in people living with CLE (95% confidence interval [CI]: 1.39, 22.27; p < 0.05) as measured by the Cutaneous Lupus Activity Investigators’ Global Assessment Revised (CLA-IGA-R) erythema score of 0-1 (clear/almost clear) at Week 16, compared to placebo (14.7% vs. 2.9%). Secondary endpoints, including the following, were not adjusted for multiplicity in Part A and therefore statistical significance cannot be demonstrated. Litifilimab was associated with rapid and continued improvement in skin disease activity with separation from placebo observed as early as Week 4 (19.3% vs. 5.5%; Δ= 13.8; CI: 1.19, 26.46), as measured by Cutaneous Lupus Erythematosus Disease Area and Severity Index Activity-50 (CLASI-50) response through Week 24 (40.8% vs. 21%; Δ= 19.8; CI: 1.46, 38.15). More participants receiving litifilimab achieved a response compared to placebo (21.7% vs 5.8%), as measured by CLASI-70 at Week 24. CLASI-50 and CLASI-70 responses are defined as 50 percent and 70 percent improvements from baseline in CLASI-A score, respectively. Additionally, 1 in 6 participants receiving litifilimab achieved a CLASI 0-3 score, defined as no or minimal disease activity, compared to placebo at Week 24 (16.3% vs 0%; Δ= 16.3; CI: 7.07, 25.62).

Litifilimab was generally well tolerated in Part A of the AMETHYST study and tolerability was consistent with the safety profile established in completed studies, including the Phase 2 LILAC study. Adverse events (AEs) were reported in 74.6% (44/59) and 64.7% (22/34) of participants receiving litifilimab and placebo, respectively, in the 24-week period. Most AEs were mild to moderate in severity. Serious adverse events occurred in 6.8% (4/59) and 2.9% (1/34) of participants receiving litifilimab and placebo, respectively.

AMETHYST is a two-part, Phase 2/3 multicenter, double-blind, placebo controlled, randomized study to evaluate the efficacy and safety of litifilimab compared to placebo. The study aims to assess the efficacy of litifilimab in participants with active subacute cutaneous lupus erythematosus (SCLE) and/or chronic cutaneous lupus erythematosus (CCLE) who are refractory or intolerant to antimalarial therapy. About Litifilimab (BIIB059)

Litifilimab (known as BIIB059), discovered and developed in-house by Biogen scientists, is a humanized IgG1 monoclonal antibody (mAb) targeting BDCA2 and is being investigated for the potential treatment of systemic lupus erythematosus (SLE) and cutaneous lupus erythematosus (CLE). BDCA2 is a receptor that is predominantly expressed on a subset of human immune cells called Plasmacytoid Dendritic Cells (pDCs). New England Journal of Medicine Publishes Positive Phase 3 VALOR Trial Results of Brepocitinib in Dermatomyositis

DURHAM, N.C., March 28, 2026 (GLOBE NEWSWIRE) — Priovant Therapeutics, a clinical-stage biotechnology company focused on developing targeted therapeutics in autoimmune disease, announced today the publication of results from the Phase 3 VALOR trial evaluating brepocitinib in adults with dermatomyositis (DM) in the New England Journal of Medicine (NEJM).

As reported in the NEJM publication, “A Phase 3 Trial of Brepocitinib in Dermatomyositis,” VALOR enrolled 241 patients across 90 sites globally and met its primary endpoint, with brepocitinib 30 mg achieving a 15.3-point greater improvement in mean Total Improvement Score (TIS) at Week 52 compared to placebo (P<0.001). This clinical improvement was observed alongside meaningful corticosteroid tapering in the brepocitinib treatment arms, with nearly twice as many patients reducing background corticosteroids in the brepocitinib 30 mg group compared to placebo. Brepocitinib 30 mg also demonstrated statistically significant and clinically meaningful improvements on all nine key secondary endpoints, with treatment effects evident as early as Week 4 and sustained through Week 52.

VALOR enrolled a broad, representative DM population that included patients with prior history of benign or malignant neoplasm and patients with multiple cardiovascular risk factors at baseline. Serious infections in the study were increased in brepocitinib 30 mg compared to placebo; these events resolved with medical management, and brepocitinib treatment was completed in most cases. Adverse events leading to treatment discontinuation occurred more frequently with placebo, as did malignancies, cardiovascular events, and thromboembolic events. The NEJM authors hypothesize in the publication that the elevated rates of these events in the placebo group reflect the baseline risks associated with dermatomyositis itself and the immunosuppressive agents — particularly systemic steroids — commonly used in its treatment. They also note the importance of the potential dual benefit of brepocitinib seen in the trial’s efficacy data – namely, reducing disease activity while simultaneously enabling substantial reductions in systemic corticosteroid exposure.

Additional Skin-Focused Analyses Presented at the 2026 Annual American Academy of Dermatology (AAD) Meeting

Additional analyses presented during the late-breaking abstract session at AAD 2026 provide deeper insight into the skin-specific and patient-reported benefits of brepocitinib beyond those reported in NEJM. These data demonstrate rapid and statistically significant reductions in itch, with an 18.9% greater proportion of patients achieving itch remission (PP-NRS ≤1) at Week 4 with brepocitinib 30 mg compared to placebo (95% CI: 5.0 to 32.9). Parallel improvements were observed in patient-reported, skin-related QoL (Skindex-16), with benefits sustained throughout the 52-week trial. Notably, among the 64% of patients with moderate-to-severe skin disease at baseline (a population often refractory to standard therapies), brepocitinib 30 mg was also associated with a 26.6% higher rate of functional skin remission compared to placebo (95% CI: 7.6 to 45.5; P=0.0060).

About the Phase 3 VALOR Study

The VALOR study was a global Phase 3 trial that enrolled 241 subjects with dermatomyositis across 90 sites. Subjects were randomized 1:1:1 to brepocitinib 30 mg, brepocitinib 15 mg, and placebo. Brepocitinib 30 mg demonstrated statistically significant and clinically meaningful improvement compared to placebo on the primary endpoint of Total Improvement Score (TIS) at Week 52. TIS is a composite endpoint of six core set measures of myositis disease activity. Benefit compared to placebo was seen as early as Week 4 and sustained at every visit thereafter through the end of the one-year double-blind treatment period. Brepocitinib 30 mg also demonstrated statistically significant and clinically meaningful improvement compared to placebo on all nine key secondary endpoints evaluated, including measures of muscle strength, skin disease activity, functional disability, and steroid tapering. More than two thirds of brepocitinib 30 mg patients achieved a Total Improvement Score of at least 40 (TIS40), twice the minimum clinically important difference. More than half achieved this TIS40 threshold while also reducing systemic corticosteroid use to ≤2.5 mg/day (prednisone-equivalent).

The VALOR trial enrolled a broad, representative DM population including patients with prior history of benign or malignant neoplasm and patients with multiple cardiovascular risk factors. Serious infections in the study were increased in brepocitinib 30 mg compared to placebo; these events resolved with medical management, and brepocitinib treatment was completed in most cases. New or recurrent malignancy, cardiovascular events, and thromboembolic events in the study occurred more frequently in the placebo arm than the brepocitinib 30 mg arm. The brepocitinib safety database across all studies includes over 2,000 patients and subjects and suggests a safety profile similar to approved JAK and TYK2 inhibitors.

The U.S. FDA has granted Priority Review to brepocitinib’s New Drug Application (NDA) and assigned a Prescription Drug User Fee Act (PDUFA) target action date in the third quarter of 2026.
