First Participants Dosed in Phase 1 Study Evaluating mRNA-1283, Moderna’s Next Generation COVID-19 Vaccine

First Participants Dosed in Phase 1 Study Evaluating mRNA-1283, Moderna’s Next Generation COVID-19 Vaccine

mRNA-1283 is being developed as a potential refrigerator stable mRNA vaccine that will facilitate easier distribution and administration by healthcare providers

CAMBRIDGE, Mass.–(BUSINESS WIRE)–Moderna, Inc. (Nasdaq: MRNA), a biotechnology company pioneering messenger RNA (mRNA) therapeutics and vaccines, today announced that the first participants have been dosed in the Phase 1 study of mRNA-1283, the Company’s next generation COVID-19 vaccine candidate.

“We are pleased to begin this Phase 1 study of our next generation COVID-19 vaccine candidate, mRNA-1283,” said Stéphane Bancel, Chief Executive Officer of Moderna. “Our investments in our mRNA platform have enabled us to develop this next generation vaccine candidate, which is a potential refrigerator-stable vaccine that could facilitate easier distribution and administration in a wider range of settings, including potentially for developing countries. We remain committed to helping address this ongoing public health emergency.”

This Phase 1 dose-ranging study will assess the safety and immunogenicity of mRNA-1283, a next-generation vaccine candidate against COVID-19. mRNA-1283 encodes for the portions of the SARS-CoV-2 spike protein critical for neutralization, specifically the Receptor Binding Domain (RBD) and N-terminal Domain (NTD). The encoded mRNA-1283 antigen is being developed as a potentially refrigerator-stable mRNA vaccine that will facilitate distribution and administration by healthcare providers. This Phase 1 study will evaluate three dose levels, 10 µg, 30 µg, and 100 µg, of the mRNA-1283 vaccine candidate given to healthy adults as a 2-dose series, 28 days apart, and one dose level, 100 µg, of mRNA-1283 given to healthy adults in a single dose. These will be compared with a 2-dose series of 100 µg of mRNA-1273, the currently authorized dose level. mRNA-1283 is intended to be evaluated in futures studies for use as a booster dose for previously vaccinated or seropositive as well as in a primary series for seronegative individuals.

About the Moderna COVID-19 Vaccine

The Moderna COVID-19 Vaccine is an mRNA vaccine against COVID-19 encoding for a prefusion stabilized form of the Spike (S) protein, which was co-developed by Moderna and investigators from the National Institute of Allergy and Infectious Disease’s (NIAID) Vaccine Research Center. The first clinical batch, which was funded by the Coalition for Epidemic Preparedness Innovations, was completed on February 7, 2020 and underwent analytical testing; it was shipped to the National Institutes of Health (NIH) on February 24, 2020, 42 days from sequence selection. The first participant in the NIAID-led Phase 1 study of the Moderna COVID-19 Vaccine was dosed on March 16, 2020, 63 days from sequence selection to Phase 1 study dosing. On May 12, 2020, the U.S Food and Drug Administration granted the Moderna COVID-19 Vaccine Fast Track designation. On May 29, 2020, the first participants in each age cohort: adults ages 18-55 years (n=300) and older adults ages 55 years and above (n=300) were dosed in the Phase 2 study of the vaccine. On July 8, 2020, the Phase 2 study completed enrolment. Results from the second interim analysis of the NIH-led Phase 1 study of the Moderna COVID-19 Vaccine in the 56-70 and 71+ age groups were published on September 29, 2020 in The New England Journal of Medicine. On July 28, 2020, results from a non-human primate preclinical viral challenge study evaluating the vaccine were published in The New England Journal of Medicine. On July 14, 2020, an interim analysis of the original cohorts in the NIH-led Phase 1 study of the vaccine was published in The New England Journal of Medicine. On November 30, 2020, Moderna announced the primary efficacy analysis of the Phase 3 study of the vaccine conducted on 196 cases. On November 30, 2020, the Company also announced that it filed for Emergency Use Authorization with the U.S. FDA and a Conditional Marketing Authorization (CMA) application with the European Medicines Agency. On December 3, 2020, a letter to the editor was published in The New England Journal of Medicine reporting that participants in the Phase 1 study of the Moderna COVID-19 Vaccine retained high levels of neutralizing antibodies through 119 days following first vaccination (90 days following second vaccination). On December 18, 2020, the U.S. FDA authorized the emergency use of the Moderna COVID-19 Vaccine in individuals 18 years of age or older. Moderna has also received authorization for its COVID-19 vaccine from health agencies in Canada, Israel, the European Union, the United Kingdom, Switzerland, Singapore and Qatar. Additional authorizations are currently under review in other countries and by the World Health Organization.

The Biomedical Advanced Research and Development Authority (BARDA), part of the Office of the Assistant Secretary for Preparedness and Response (ASPR) within the U.S. Department of Health and Human Services (HHS) is supporting the continued research and development of the Company’s COVID-19 vaccine development efforts with federal funding under contract no. 75A50120C00034. BARDA is reimbursing Moderna for 100 percent of the allowable costs incurred by the Company for conducting the program described in the BARDA contract. About Moderna

In 10 years since its inception, Moderna has transformed from a science research-stage company advancing programs in the promising-but-still-unproven field of messenger RNA (mRNA), to an enterprise with its first medicine having treated millions of people, a diverse clinical portfolio of vaccines and therapeutics across six modalities, a broad intellectual property portfolio in areas including mRNA and lipid nanoparticle formulation, and an integrated manufacturing plant that allows for both clinical and commercial production at scale and at unprecedented speed. Moderna maintains alliances with a broad range of domestic and overseas government and commercial collaborators, which has allowed for the pursuit of both groundbreaking science and rapid scaling of manufacturing. Most recently, Moderna’s capabilities have come together to allow the authorized use of one of the earliest and most-effective vaccines against the COVID-19 pandemic.

Moderna’s mRNA platform builds on continuous advances in basic and applied mRNA science, delivery technology and manufacturing, and has allowed the development of therapeutics and vaccines for infectious diseases, immuno-oncology, rare diseases, cardiovascular diseases and auto-immune diseases. Today, 24 development programs are underway across these therapeutic areas, with 13 programs having entered the clinic. KeyedIn Announces Updates to its Project Portfolio Management Solutions to Simplify Agile Adoption and Portfolio-Level Decision-Making

MINNEAPOLIS, March 15, 2021 (GLOBE NEWSWIRE) — KeyedIn, a leader in Agile Portfolio Management, today announced a variety of updates to its cloud-based project portfolio management (PPM) solutions designed to help project management offices (PMOs) simplify Agile adoption and leverage portfolio reporting to make better data-driven decisions.

KeyedIn has offered innovative Kanban-style Agile planning at the portfolio level for several years and is now extending that capability to the task level. Both Kanban-style tools – one for portfolio projects and now another for tasks – enable PMOs and enterprise PMOs (ePMOs) to more easily incorporate Agile projects and tasks alongside traditional or custom work methodologies, to give portfolio leaders comprehensive visibility into all project-based work. The new Task Board also allows non-IT Agile projects to be run solely in KeyedIn without the need for additional Agile tooling.

“The ability to manage any work methodology within the same portfolio is a core tenet of KeyedIn’s Agile Portfolio Management strategy,” said Matt Muldoon, Chief Product Officer for KeyedIn. “The Task Board gives portfolio leaders flexibility in how they track Agile projects – either natively in KeyedIn or through integration with Jira or similar tools. Getting that complete picture is what enables KeyedIn customers to place the right bets, turn quickly and delivery faster.”

With this new feature, KeyedIn allows users to seamlessly switch between a Gantt chart planning tool and the Task Board, depending on which best fits the work in process. On the Task Board, each item moves through standardized progress stages so teams can track and update what’s in process at a glance and identify bottlenecks.

In addition, the updates to KeyedIn’s PPM solutions include several reporting enhancements, including the ability to define and execute PowerPoint reports for the first time. This format is especially useful for customer-specific status reports and “packs” that contain summaries of many projects for senior stakeholder presentation. Users can also now embed iFrame widgets into KeyedIn dashboards, to bring data from third-party products into the KeyedIn ecosystem for integrated visibility.

“We’ve invested a tremendous amount of development time and resource into giving KeyedIn the best self-service reporting tools among any PPM product,” Muldoon said. “The project and portfolio data stored in a PPM tool holds the key to improving forecasting, prioritizing the most valuable projects and identifying problems early – but only if portfolio leaders can get to it and filter it to their immediate needs. KeyedIn makes that possible.”

These new enhancements are part of the version 7.3 release of KeyedIn’s suite of PPM products. The Kanban-style Task Board will be available in April 2021 to KeyedIn Enterprise (KIE) and KeyedIn Projects (KIP) customers. The reporting enhancements are available across all KeyedIn product editions, including KeyedIn Adaptive Portfolios (KAP). Bombardier’s Demonstration Aircraft Operations Team Earns IBAC’s Highest Recognition for Enhanced Safety Culture

  • Stage 3 IS-BAO certification validates Bombardier’s vigour in developing and maintaining an effective safety culture
  • Bombardier first achieved Stage 3 IS-BAO safety rating in 2018; renewal further highlights ongoing efforts to exceed all safety protocols
  • Bombardier’s Safety Standdown event and monthly Safety Talks highlight industry-wide commitment to ongoing knowledge-based safety training

MONTREAL, March 15, 2021 (GLOBE NEWSWIRE) — Bombardier announced today that its demonstration aircraft operations team has once again earned the International Standard for Business Aircraft Operations (IS-BAO) Stage 3 accreditation for its ongoing commitment to the implementation of a robust safety culture.

Stage 3 designation is the highest level of recognition issued by the International Business Aviation Council (IBAC), and this year’s award marks the second consecutive time that the Bombardier demonstration aircraft team has earned this designation. The team first achieved Stage 3 certification in 2018, IS-BAO’s highest safety rating for developing and maintaining an effective Safety Management System (SMS) for its demonstration aircraft. The designation, given out every three years, continues to shine a light on the organization’s overall commitment to an outstanding safety culture for its customers and employees.

“Bombardier is committed to ensuring all levels of its operations meet the highest safety standards, and this recognition from IBAC aptly reflects the high safety standard displayed by our demonstration aircraft operations team,” said Andy Nureddin, Vice President, Customer Support, Bombardier. “Our goal is to meet and exceed safety protocols and professionalism throughout all of our operations, and we will continue to ensure our SMS program exceeds all industry safety standards.”

Based in Hartford, Connecticut, Bombardier’s demonstration aviation team prides itself in delivering the safest flying environment while delivering an amazing experience to its customers. The team is dedicated to achieving a higher level of corporate responsibility and professionalism in the aviation industry, and aims to provide expertise, guidance, mentoring and creative support solutions to its customers. In addition to implementing stringent measures to attain Stage 3 status, the crew also enhances the company’s safety culture by vetting charter operators for crew hours, holding monthly safety committee meetings and implementing stringent Covid-19 procedures for crew and passengers.

Officially recognized as the global safety standard for business aviation by the International Civil Aviation Organization, IS-BAO helps operators apply industry best practices by challenging them to review and compare their safety-related policies, processes and procedures, and then make improvements, elevating them to the worldwide standard for business aviation.

This commitment to the highest safety protocols extends beyond this organization. Bombardier is a longtime leader and steward of the tremendously important air safety portfolio. With its strong engagement in drafting ICAO’s Annex 19 on Safety Management Systems and its ongoing contribution to international air safety and accident organizations, Bombardier is a true leader in developing a robust safety culture. The company also supports numerous safety workshops and seminars with aircraft manufacturer and business jet operator associations worldwide.  Additionally, Bombardier organizes and hosts the awarding-winning Safety Standdown – recognized as one of the top annual safety gatherings in civil aviation, celebrating its 25th anniversary in 2021.

The Bombardier demonstration aircraft team is a steward of our annual Safety Standdown event. Safety Standdown provides aviation professionals with essential knowledge-based aviation training and learning. Held in concert with a series of comprehensive monthly Safety Talks, Standdown’s goal is to foster a community of aviation professionals who are committed to lifelong learning and to disseminating higher standards of safety and professionalism throughout the industry.

Researchers Unveil a New Theory Explaining Long-term Pain Relief

Stimulation of peripheral nerves warrants consideration early in the care continuum

CLEVELAND, March 15, 2021 (GLOBE NEWSWIRE) — A novel theory recently published in the Journal of Pain Research proposes mechanisms by which a 60-day treatment using percutaneous peripheral nerve stimulation (PNS) may recondition the central nervous system (CNS) to restore normal pain processing and provide long-term pain relief for patients. This short-term treatment may replace the need for permanently implanted systems in many patients.

Chronic pain may occur due to an imbalance in nerve signal processing within the CNS as a result of reduced healthy inputs coming from within the region of pain. The new theory proposes that increasing healthy neural inputs by stimulating peripheral nerves can recondition the CNS and restore healthy pain processing. Pain relief following a 60-day peripheral nerve stimulation treatment has been reported by some patients to endure for several years or more.

In the case of low back pain, for example, lower back muscles are often inhibited, contracting less and consequently conveying fewer healthy messages to the cortex. In this case, PNS can be used to induce comfortable cycling contractions within the region of pain. Restoring the healthy, non-painful signals may normalize central processing resulting in the elimination or reduction of pain. Similar indications of central reconditioning have also been demonstrated when a 60-day PNS treatment is applied to the nerves of patients with post-amputation pain, in this case without causing a muscle contraction.

According to Dr. Timothy Deer, President & CEO of The Spine and Nerve Center of the Virginias and lead author of the publication, “The theoretical framework for central reconditioning provides a significant conceptual advancement in regard to how a short-term treatment may produce the long-lasting results that many of my pain management colleagues and I are seeing on a routine basis following the SPRINT PNS System’s 60-day treatment.”

“The clinical implications of this theory support the notion of preferentially using a short-term PNS treatment early in the care pathway as a means of helping patients, payers and pain management physicians avoid the more invasive and costly permanent implantation of neurostimulation systems that have been in routine use over the past half-century,” said co-author Peter Staats, MD, MBA, SPR Therapeutics’ Chief Medical Advisor and President of the World Institute of Pain.
